Support has said numerous times in the past they will not change a guild leader. The only way for leadership to change is to have the current leader pass leadership or start a new guild.The only possible way, ingame, to change the guildmaster is the guildmaster him-/herself by choosing someone or leaving the guild (gm passes over to player with the next highest rank).
So, the only way for you would propably be the support. Maybe because of a good reason they can make you guildmaster.

Contrary to what others have said, I'd recommend contacting Support and ask them to give you the guild leader's spot. Give them as much detail as you can, including the server, guild name, current inactive guild leader's name, how long he/she has been inactive (insofar as you can determine), and so on. It can't hurt to try and might just help. Support has transferred guild leadership in the past, but not without a very good reason.
If you do this please be patient. It can take a fair amount of time for Support to do the investigation.
